RAD-140 or Testolone, is a powerful orally active anabolic in a group of chemicals known as SARMS (Selective Androgen Receptor Modulators). RAD-140 appears to have a high selectivity for targeting bone and lean muscle tissue over other tissues such as organs. Although it does appear to activate androgens outside of these tissues, it is comparatively less than with anabolic steroids or natural testosterone. Testolone has an anabolic androgenic binding ratio of 20 to 1; the powerful SARM, LGD-4033, has a ratio of 10:1.
Being presented in 2010, RAD-140 is a whole lot newer to the scene than many other chemicals in the selective androgen modulator class. The compound works by selectively targeting the androgenic receptors in specific tissues. Comparatively, anabolic steroids have no selectivity by tissue and this can generate a large list of serious and long-term side effects that may be much less with SARMs. Theoretically, substances in the SARM class have lower risks of negative side effects.
The FDA warns of long-term risks unknown in SARM use and also warns of a potential for increased heart attack and stroke. To date, no human clinical research has been carried out on RAD-140 but there is a range of animal studies showing promising results and safety. There are many anecdotal reports online of bodybuilders using this substance to increase lean muscle mass alone or in combination with other anabolic substances.
Clinical Research and Human Investigation
Pre-clinical trials for RAD-140 have been performed on animals to date. Therefore the compound has not yet officially received clearance for human clinical research trials.
In an animal rat study, RAD-140 demonstrated no risk of increased prostate size. An enlarged prostate is a common side effect of nearly all anabolic steroids. Further investigation is needed to investigate the effects of testolone on the human prostate.
Testolone is a powerful SARM with an anabolic bs androgenic ratio of 20:1. Comparatively, LGD-4033 has a binding ratio of 10:1 and natural testosterone 1:1. LGD-4033 has untaken phase 1 human trials. In phase 1, LGD-4033 showed limited side effects and anabolic properties that are consistent with SARM theories. In theory, testolone should show similar results to LGD with even fewer, androgenic side effects. The LGD-4033 human study showed impairment of natural testosterone production but levels quickly returned to normal with no hormonal drug therapy needed. RAD-140 should show a similar safety profile but more research is needed to draw any conclusions about either SARM.
In a study performed on rats to test the effectiveness of Testolone against age-related cognitive decline and dementia; results implicated improved cognitive functioning. As much of the scientific world has already noted, aging of the male gender and the inevitable fall of testosterone production has the negative effect of heightening men’s likelihood of succumbing to illness, viruses or even long-standing physical pains.
One safety concern to seriously consider with any synthetic substance consumed by humans is the risk of allergic reaction. Although usually rare, both synthetic and natural drugs can cause rejection by the body. An allergic reaction usually varies in severity, however, this risk with RAD-140 and related substances cannot be ruled out as a serious risk factor. No current human research investigating SARMs and allergic reactions exists at this time.
SARMs like RAD-140 have full attention in the medical community due to their lack of androgenic side effects and high selectivity for muscle tissue. Some of the most promising conditions that may be treated with these substances include: Testosterone Replacement Therapy, various endocrine disorders, osteoporosis and muscle wasting syndromes. With the limited data available, SARMs like RAD-140 show promising targets to replace anabolic synthetic testosterone.
Additionally, SARMs show a relative lack to increase dihydrotestosterone, a male sex hormone that causes masculinization such as deepened voice and hair growth. For obvious reason, RAD-140 and other anabolic androgen receptor modulators may be more attractive to a woman than with standard steroidal treatments used in medicine. Increases in DHT activity is also believed to play a role in balding in adults so this is another possibility that may make the SARM class a more appealing option.
Side-effects of RAD 140
Published research isn’t available on the side effects of RAD-140 in humans. On the FDA website, they issued a recent blanket warning about SARMs and state the drug class increases the risk of heart attack and stroke. Steroids also increase this risk although it is unsure how they compare in this regard. The FDA does not make it clear in the post the mechanisms that increase these risks or any figures, data or study in this area. In addition to these serious risks, the FDA states SARMs can cause liver toxicity. References to liver toxicity were in reference to those supplementing SARMs in the past. The FDA warning did no address whether or not these past events involving liver toxicity were caused by a standalone SARM or in products containing one or more as ingredients.
As an interesting side-note, one rat study showed RAD-140 to be neuroprotective in cases of cell death brought on by a stroke. It is unclear how these claims tie into the FDA blanket statement that SARMs increase the risk of stroke.
In theory, SARMs like RAD-140 should carry a lower risk of heart complication, stroke and liver impairment. Human research on several SARMs has shown normal liver function with little to no impairment in healthy groups. Steroids on the other hand, especially ones taken orally, have shown determinantal effects to the liver. Oral steroids are especially harmful due to how they are modified so that the liver cannot eliminate them quickly. Oral SARMs, in general, do not have this same issue. Regardless, more study needs to be invested to draw any conclusions or comparisons on the safety of SARMs on essential organ function.
There is no safe way for increasing androgenic activity through pharmacological data. However, on paper, testolone, appears to have much fewer side effects than steroidal drugs. The side effect profile is likely to be similar to LGD-4033.
